PTEN hamartoma tumor syndrome is an Autosomal dominant clinical disorder associated with mutations of the PTEN or phosphatase and tensin homolog gene on chromosome 10.

Cowden syndrome is the most common phenotype, with an estimated incidence of 1 in 200,000-250,000 people.
