Daraxonrasib

Daraxonrasib is an investigational, once-daily oral targeted therapy developed by Revolution Medicines for cancers driven by RAS mutations.

It is a ( “RAS(ON)” inhibitor that works by blocking mutated RAS proteins (such as KRAS) across multiple variants, which historically were considered undruggable.

The drug has shown unprecedented results, notably doubling median overall survival to approximately 13.2 months compared to 6.7 months with standard chemotherapy in patients with previously treated metastatic pancreatic cancer.

While most targeted therapies only work for specific gene variants, daraxonrasib works as a multiselective inhibitor that turns off various overactive KRAS proteins to stop tumor growth.

Side Effects: While not completely devoid of side effects, it is considered much more tolerable than standard cytotoxic chemotherapy, allowing for better quality of life.

Common side effects include rash, diarrhea, nausea, and mouth sores.

Other Cancers: In addition to pancreatic ductal adenocarcinoma (PDAC), clinical trials are actively studying its efficacy in treating other RAS-mutated solid tumors, such as non-small cell lung cancer (NSCLC) and colorectal cancer.
