Vusolimogene oderparepvec-wtpg

Vusolimogene oderparepvec-wtpg (brand name Tudriqev) is a genetically modified oncolytic viral immunotherapy.

It is built from a modified herpes simplex virus type 1 (HSV-1).

Cancer Killing: It selectively targets, infects, and replicates inside tumor cells—causing them to burst while leaving healthy cells unharmed.

It expresses human GM-CSF and a fusogenic glycoprotein to trigger a systemic immune response against the cancer.

This type of treatment is called an “oncolytic viral therapy”—meaning a virus-based treatment that kills cancer cells.

Approval for Tudriqev combined with nivolumab for adults with advanced unresectable or metastatic cutaneous melanoma progressing after anti-PD-1 therapy.

Administration Given as an intratumoral injection every 2 weeks for 8 doses, starting at 10⁶ PFU/mL and increasing to 10⁷ PFU/mL, combined with intravenous nivolumab starting at Week 3.

It is used with nivolumab to treat adults with advanced melanoma of the skin that cannot be removed surgically and has progressed despite treatment with a PD-1–blocking immunotherapy.

A healthcare professional injects Tudriqev directly into suitable tumors every two weeks for eight doses.

Nivolumab is given separately by intravenous infusion, beginning at week 3. Tudriqev itself is not administered intravenously.

The IGNYTE trial, which enrolled 140 adults whose advanced melanoma had progressed after prior anti-PD-1 treatment.

The objective response rate—the proportion whose tumors shrank enough to meet the study’s response criteria—was 24.2%.

Among those who responded, the median duration of response was 14.1 months.

The study was single-arm, meaning it did not have a randomized comparison group.

Common adverse reactions include fatigue, fever, chills, nausea, diarrhea, muscle or joint pain, and injection-site reactions. Important warnings include accidental exposure to the modified virus, herpes infection or reactivation, complications from tumor injections, and immune-mediated adverse events.

Handling and exposure precautions matter because this is a live, modified herpes-virus treatment.
