Src family of protein kinases

Linked to signaling to initiate cell growth factors, integrin, cytokine receptors on cell surfaces and can effect downstream signaling cascades.

Can help coordinate cancer cell growth, survival, invasion and angiogenesis.

Can cooperate with multiple receptor tyrosine kinases, such as EGFR to modulate, signaling, transform cell and promote tumor growth.

Activates downstream signaling pathways including Ras/Raf/MAPK, P13K/Akt and STATs.

VEGF and IL-8, angiogenic growth factors are downstream targets for c-Src.

Inhibitors of Src can inhibit angiogenesis.

Overexpressed in prostate cancer and other malignancies.

Fyn is the most overexpressed Src family of protein kinases in prostate cancer, with increased expression with metastatic disease. And with the transition to castrate resistant disease.

Fyn is associated with T-cell activation and neuronal development.
