Trametinib

Trametinib (brand name Mekinist) is a targeted cancer therapy known as a MEK inhibitor.

It works by blocking specific proteins (MEK1 and MEK2) that signal cancer cells to multiply and spread.

Trametinib is primarily used to treat cancers that have a specific genetic mutation called BRAF V600E or V600K.

It is often prescribed in combination with another drug, dabrafenib (Tafinlar), for the following conditions:

Melanoma: Used for metastaticcancer that cannot be removed by surgery, or as an adjuvant treatment to prevent recurrence after surgery.

Non-Small Cell Lung Cancer (NSCLC): For metastatic lung cancer with the BRAF V600E mutation.

Thyroid Cancer: Specifically for locally advanced or metastatic anaplastic thyroid cancer.

Solid Tumors: For adults and children (6+ years) with BRAF V600E-mutant solid tumors that have progressed after other treatments.

Glioma: For children (1+ year) with low-grade glioma.

In many cancers, the “MAPK” signaling pathway is overactive, telling cells to grow uncontrollably.

Trametinib binds to and inhibits MEK1 and MEK2, two enzymes in this pathway.

By shutting down this signal, the drug causes cell cycle arrest and slows the growth of the tumor.

It is an oral medication taken once daily as a tablet or oral solution.

Common Side Effects: Fatigue, fevers, prash, nausea, diarrhea, and swelling.

Monitoring: Because it can affect liver enzymes, regular blood tests are typically required during treatment.
