Relacorilant (Lifyorli)

Relacorilant (Lifyorli) is an oral, selective glucocorticoid receptor antagonist approved for use with nab-paclitaxel to treat specific advanced ovarian, fallopian tube, or primary peritoneal cancers.

It specifically binds to the glucocorticoid receptor to block cortisol activity without impacting other hormone receptors.

Sensitization: Reduces cortisol-driven survival signals in tumor cells, increasing sensitivity to chemotherapy.

It helps counter tumor resistance to cytotoxic treatments like taxanes.

DosageRegimen: 150 mg taken orally on the day before, day of, and day after each nab-paclitaxel infusion.

Chemotherapy: Typically, 80 mg/m² nab-paclitaxel is administered on days 1, 8, and 15 of a 28-day cycle.

Continued until disease progression or toxicity.

Side Effects Common adverse reactions include neutropenia, anemia, fatigue, and nause

Relacorilant added to nab-Paclitaxel (Abraxane) improves overall survival versus Abraxane alone, in patients with platinum resistant ovarian cancer.
