Capivasertib (Truqap) is an oral AKT inhibitor that blocks signaling pathways responsible for cancer cell growth.

It is FDA-approved for treating certain advanced cancers identified through molecular testing:

Breast Cancer: Used with fulvestrant for HR-positive, HER2-negative, locally advanced/metastatic cases with PIK3CA, AKT1, or PTEN alterations after endocrine therapy.

Prostate Cancer: Paired with abiraterone/prednisone for PTEN-deficient, metastatic castration-sensitive (mCSPC) disease.

Dosage and AdministrationThe medication is typically taken orally at a dose of 400 mg twice daily.

It follows an intermittent schedule—4 days on, 3 days off weekly—to manage side effects, and tablets must be swallowed whole.

Side Effects include diarrhea, rash, nausea, and hyperglycemia.

Due to risks like embryo-fetal toxicity, close monitoring by a healthcare provider is required.
